HeLa cells were incubated at 37 oC with 1X of SYBR Green I, MxSafe and MxSafe Plus,respectively. Images were taken
following incubation for 5 min (panel A) and 30 min (panel B), respectively.SYBR Green I entered into cells rapidly as ev
-ident from the bright green nuclear staining. However, MxSafe and MxSafe Plus were unable to cross cell membranes
as shown by the lack of any fluorescence staining.
MxSafe Plus is an ultra sensitive, extremely stable and environmentally safe fluorescent nucleic acid dye designed to replace
the highly toxic ethidium bromide (EB) for staining dsDNA, ssDNA or RNA in agarose gels or polyacrylamide gels. MxSafe Plus
is far more sensitivethan EB without requiring a destaining step. MxSafe Plus and EB have virtually the same spectra, so you
can directly replace EB with MxSafe Plus without changing your existing imaging system.
MxSafe Plus is a very stable dye. Store 10,000X solution and dilute solutions of MxSafe Plus at room temperature,
protected from light. Dye precipitation may occur at lower temperatures, resulting in lower signal or the
appearance of precipitate on the surface of the gel. If this occurs, heat the solution to 45-50oC for two minutes
and vortex. MxSafe Plus is stable for at least one year from the date it is received.
